Uses of Tafero Tablet


Tafero  25mg is a brand name of Tenofovir alafenamide. It is an antiretroviral drugs and involves in nucleotide reverse transcriptase inhibitor(NRTI). In adult is used  for the treatment of Chronic HBV infection with compensated liver disease and HIV infection

Side effects of Tafero Tablet

Common side effects:

Pain in black ; cough and headache

Side effects not known:

  • Abdominal or stomach discomfort

  • bloody urine dark urine

  • lack of appetite

  • decreased frequency of urine

  • hypertension

  • increased thirst

  • light-colored stools

  • cramping nausea and vomiting

  • fullness sleepiness

  • swelling of the face, fingers, or lower legs

How to use Tafero Tablet


Before starting of treatment Tafero 25mg, patient should screening for  HIV-1 infection. Tafero alone should not be given for patients with HIV infection.

In adult Tafero is used for chronic HBV infection which the drug  contains Tenofovir Alafenamide 25 mg single tablet taken oral route once in a day with food.

No dosage adjustment required for renal and hepatic impairment.

 Overdose: If Tafero overdose occurs, check the patient for probability of toxicity and provide supportive measures or seek to emergency medical department or call poison helpline immediately

How to Tafero Tablet works:


 Tenofovir alafenamide is an antiviral drug against the hepatitis B virus and also used in some HIV infections

 Tenofovir  Alafenamide (Tafero 25mg) is a inhibitor of Nucleotide reverse transcriptase that fight against viral infection . which act as an Phosphonamidate prodrug, then Tenofovir is hydrolized in vivo and then phosphorylated to the active metabolite to form Tenofovir diphosphate. Drug will prevents the  reproduction of HIV virus in human cells and then stops  the HIV virus from producing new viruses and a novel ester prodrug of the antiretroviral tenofovir, use to treat  HIV infection and chronic hepatitis B.Tafero 25mg cause fast chain termination and interferes with proviral DNA 27 transcription.

Absorption: Tenofovir alafenamide Peak plasma concentration is approx 0.5hrs after the dose. Food effect: Tenofovir alafenamide  with a high fat meal increase AUC compared with administration in fasted state

Distribution: Plasma protein binding is 80%

Metabolism : Only minimal metabolized by CYP3A

Elimination : Excreted by kidneys, 32% of dose in feces and<1% excreted in  urine. Half life is 0.5hrs.

Expert Advice for Tafero Tablets


  • Tafero 25 mg co administration of antiretroviral agaents or alone use will cause severe hepatomegaly including fatalities and lactic acidosis.
  • Individual treatment of  Tafero should not be used for treatment of HIV infection causes risk of producing of HIV-1 resistance.
  • While taking Tafero 25mg along with renal impaired function or nephrotoxic agents will increase risk of developing renal related side effects.
  • If patient discontinued of Tafero 25mg drug it will lead to severe acute exacerbation of HBV infection. Hence liver function should monitor after several month discontinue of drug.


Special Warnings of Tafero Tablets



Alcohol


On treatment with Tafero 25mg drinking alcoholic beverages is safe or not depends upon number of factors involves the patient receiving drugs. Alcohol may inhibits some action of hepatitis drugs work. So patient should discuss with doctor about any adverse reaction of drinking during treatment.

Pregnancy


 Category B : While Tafero 25 mg during pregnancy ,no malformation, fetotoxicity or neonatal toxicity, Tafero referred  for only limited number of pregnant women. In  animal reproduction studies have shown negative risk to the fetus and there no sufficient and well – tolerated studies in pregnant women. This drug harms the unborn baby.

Lactation


During Tafero 25mg treatment, breast feeding is avoided for the safety of the newborn baby. This drugs may included in breast milk in high levels and probably harm the baby. Check with the doctor whether breast feeding should stop temporarily or permanently while treatment

Driving


If dizziness, sleepy, tried or low alertness on drive happens then do not drive until you feeling well.



Kidney


Tafero 25mg treatment of patients with mild moderate, severe renal impairment has no dose adjustment and the drug is not recommended in patients with end stage renal disease. Consult with your doctor for further details.

Liver


Patient with decompensated hepatic impairment is not recommended with Tafero treatment, because safety and efficacy is not been established. No adjustment is required. Consult with your doctor



Missed Dosage


As soon as you remember take the missed dose. If time for next dose then skips the missed dose and follow the routine scheduled. Do not take more than one tablet per day. Please consult your doctor for changes in dose or new schedule for replace of missed dose.
